In Arizona in the late 1800s, infamous outlaw Ben Wade and his vicious gang of thieves and murderers have plagued the Southern Railroad. When Wade is captured, Civil War veteran Dan Evans, struggling to survive on his drought-plagued ranch, volunteers to deliver him alive to the "3:10 to Yuma", a train that will take the killer to trial.

Stodge City is in the grip of the Rumpo Kid and his gang. Mistaken identity again takes a hand as a 'sanitary engineer' named Marshal P. Knutt is mistaken for a law marshal. Masquerading as a parson and a drifter, Sandy Hopkins (Raymond Hatton) and Nevada Jack McKenzie (Mack Brown) come to the aid of the beleaguered residents of Goldville, a small ranching community being terrorized by greedy saloon keeper Ace Benton (Kenneth MacDonald) and his gang of cutthroats. Unbeknownst to the citizenry, the railroad is planning to build tracks through town and Benton is attempting to secure the land by scaring off the settlers.
